APPLICATION,This method is applicable to the sampling and examination of fig paste for insect and mites and other extraneous,material to determine compliance with Sections 4, 5 and 7 of the Food and Drugs Act. This method replaces,ExFHPB-6, dated April 1996.,2. DEFINITION OF TERMS,A lot is defined as that amount (volume, weight, etc.) of fig paste which is produced, stored and/or shipped under,conditions as nearly uniform as possible, consisting of not more than one variety, grade or type of product from a,single identifiable source.,3. COLLECTION OF SAMPLES,3.1 Scrutinize the entire lot for live infestation. If live infestation is found, do not sample until after fumigation,or other effective treatment.,3.2 Obtain eight sample units selected at random from the lot of at least 200 g each using appropriate,sampling equipment and containers. Eight sample units constitute a sample.,3.3 Each sample unit must be kept separate and labelled 1, 2, 3, etc. Complete information respecting the lot,size, country of origin, exporter, importer and product identification should be recorded and should,accompany the sample.,4. MATERIALS AND SPECIAL EQUIPMENT,1) Balance,2) Wildman 2 L trap flasks (AOAC 945.75B (4)),3) Filtered or distilled water,4) Stirrer hot plate,5) Magnetic stirring bar (1 x 5 cm recommended),6) Deodorized kerosene,ExFHPB-6,February 2006,-2-,7) Beakers (0.4 - 1.0 L recommended),8) Large funnel (~ 15.0 cm top diameter),9) U.S. standard sieves, No.20, No.140 and No.230,10) Wash bottles,11) Suction filtration apparatus. Use either a Hirsch or Buchner funnel. For both funnels, the filter paper (S&S,#8 ruled) should be larger than the funnel plate so the paper forms a cup in the funnel. Moisten the filter,paper and make sure it gives good suction before filtering.,12) Clearing agent. A glycerol-alcohol (95%) mixture (1:1) is recommended.,13) Petri dishes,14) Alcohol. Use 95% ethanol or 99% isopropanol,15) Stereoscopic microscope (10-70x),16) 10% Na4 EDTA solution,17) pH paper - broad range,5. PROCEDURE,The examination shall be carried out in accordance with the following instructions.,5.1 Analytical Units,5.1.1 Break off portions of fig paste in a random fashion from a sample unit and weight out 100 g. This,100 g amount constitutes an analytical unit.,5.1.2 Repeat Step 5.1.1 for each of the remaining seven sample units.,5.2 Isolation : Light Filth,5.2.1 Transfer a 100 g analytical unit of fig paste to a 2 L trap flask, breaking up fig paste only enough,to enter flask.,5.2.2 Dilute mixture to 900 mL with hot water. Bring contents to a boil and boil $ 20 min with occasional,stirring until all lumps are disintegrated.,5.2.3 Allow mixture to cool undisturbed to between 20 and 25oC.,5.2.4 Add 50 mL of kerosene and a magnetic stirring bar.,5.2.5 Slowly bring stirrer to maximum speed that does not produce audible or visible splashing or a,permanent emulsion. All of the material should be in suspension and a portion of the stirring bar,should be visible at the bottom of the vortex. Stir magnetically 5 min.,5.2.6 Slowly add deaerated water down trap rod, maintaining top of stopper just above kerosene layer.,When stopper is almost fully raised, continue adding water until kerosene layer passes the stopper,and the bottom of kerosene layer is approximately 1 cm above top of fully raised stopper.,5.2.7 Swirl stopper through lower portion of trap flask so as to suspend settlings.,ExFHPB-6,February 2006,-3-,5.2.8 Let mixture stand 30 min with intermittent stirring of bottom layer during first 20 min.,5.2.9 Spin rod slowly to remove sediment that may be on top of stopper and at the same time gently,raise rod to highest position. While at this position, rinse rod and neck of flask with water. Hold,stopper in place and trap off by pouring liquid into beaker.,5.2.10 Rinse out material on rod and in neck of flask with water and add to beaker.,5.2.11 Lower rod gently without losing any liquid by sudden splashing especially when stopper is stuck,within neck of flask.,5.2.12 If amount of seeds and/or other plant material that has been trapped off is excessive such that it,may subsequently interfere with examination of filter paper, the trapped-off material may be sieved,……
